Cannonball Adderley étudie au lycée de Tallahassee (Floride) où il apprend la flûte, la trompette, la clarinette et l'alto, avant de diriger un orchestre scolaire à Fort Lauderdale, il y reçoit le surnom de « Cannibal » transformé plus tard en « Cannonball ».
Puis il enseigne la musique et dirige plusieurs formations locales en Floride. En 1955, lors d’un voyage à New York, il se fait remarquer par le label Savoy qui le signe et lui propose de s’intaller à New York. Il commence alors à jouer dans les jam sessions et dans des enregistrements de Kenny Clarke et de son frère Nat Adderley, puis sous son propre nom. Cannonball Adderley est alors considéré comme « le nouveau Charlie Parker Il fonde avec son frère Nat Adderley, un quintet, et un sextet en 1959. En 1958, il accompagne de Miles Davis, pour enregistrer Milestones, Porgy and Bess (1958), Kind of Blue (1959). Il invite Miles Davis sur son album Somethin' Else sorti en 1958, qui restera l'une des rares apparitions phonographiques de Miles en tant que sideman ! À partir de 1960, il enregistre sous son propre nom et sous celui de son frère, en compagnie des pianistes Bobby Timmons, Hank Jones, Bill Evans, Wynton Kelly, Victor Feldman, des bassistes Paul Chambers et Sam Jones, des batteurs Albert Heath et Louis Hayes. Dès 1968, en compagnie des pianistes Joe Zawinul et George Duke, Cannonball Adderley se tourne vers une musique bluesy, funk et soul, qui marquera le début des évolutions commerciales du jazz.
